No, they are direct competitors and specifically are against one another. You can't read nook books using a Kindle or Kindle app and you can't read Kindle ebooks using a nook or nook app. You can't download books for the Kindle using a nook and viceversa.

As a side note, the nook color can be hacked to turn it into an android tablet, but it does require a good degree of technical knowledge so I don't recommend it because it voids the warranty and nook color is a pricey gadget. Not worth the risk to do so unless you truly know your way around this type of gadget.
References :
Kindle uses the Amazon Kindle format and the Nook uses the Barnes & Nobles format. Neither wants you to use the other store to buy your ebooks.

Now, there are other format that you can load into both, but each ereaders wants you to shop at their own store.
References :
Avid Kindle user (1st & 3rd gen)

Most for-purchase books are protected by DRM (Digital Rights Management) systems that are particular to the ereader, so, no, you can't count on being able to buy B&N ebooks for your Kindle, or Amazon ebooks for your Nook. There are lots of places, however, which offer free books that will work on most ereaders. Sometimes its a single format that most support (EPUB, TXT, PDF, etc); sometimes multiple ebook formats are listed per book.

The sites listed here have for-purchase ebooks and free ebook downloads:

http://www.howtodecide.com/ebook-download/

Also, if you master a good conversion tool such as Calibre,

http://calibre-ebook.com/

you'll gain access to more formats than just your ereader's native ebook file formats. Calibre will not, however, overcome DRM of protected for-purchase books.
References :
it all depends on the format of the ebook. i believe most ebooks are either in .pdf or .epub

i do not believe the kindle reads .epub books.
References :
No, you can't resell any ebooks you purchase unless you actually own the rights to do so, and just buying an ebook from an ebook store does not give you that right. It's one of the downsides of electronic v. print.

The main advantage of ebooks v. print are:

1. Instant gratification if the ebook is released at the same time as the print version, provided you have access to ebook store(s) that sell the ebook. It's a matter of a couple of clicks and a download.

2. The ability to carry hundreds and even thousands of books with you at a given time. If your ebook reader has a slot for an SD card, you can pretty much carry a whole library with you at all times.

3. Related to the above, space saving convenience.

There are quite a number of things pertaining to ebooks that still need to be sorted out, i.e. pricing, geographical restrictions, DRM issues, etc. Personally, since I am in the US, I don't have to deal with geographical restrictions, I don't buy any e-books that are more expensive than its equivalent mass market paperback, and I strip the DRM so that I can not only read my purchases in whatever device I want, but also convert them to multiple formats. Again, it's pretty much about the convenience, but e-books have a ways to go still, particularly if you live overseas, or you prefer to resell your books once you are done reading them.
References :
One looses the touch, feel and smell of paper, the holding of an item with the dimension, the ability to set it down and not to be concerned about the battery dying or if running on ac power, the thunder and lighting storm outside. And if you spill your coffee or your drink, the book will dry out and may still be readable, while the electronic tool to read the book is shocked to hell.

Can you tell....my opinion is books are so much more enjoyable as paper bound than e-bound.
